Why does a working microphone appear unavailable?

Browser permission, operating-system access, a different selected input or another application can affect the result. Check these before assuming hardware failure.

Does one successful test cover every application?

No. Another application may use a different device, input level or permission setting. Test in the application where the device will be used as well.
